Developing a sensitive, ecological, and economic HPLC method for the concurrent quantification of KOLCHICINE and KHELLIN, used in bulk and sachet pharmaceuticals

Abstract:
Establishing adequate stability-indicating research has garnered a lot of interest lately in drug testing. We have designed and verified a thorough, kinder-to-the-environment stability-indicating RP-LC strategy to detect the levels of colchicine (COL) and khellin (KHE) in sachet dosage forms. Colchicine and khellin underwent a number of stress tests in accordance with the International Conference on Harmonization's (ICH) criteria. The column with C-18 was used for analyzing both drugs at the ambient temperature after the isocratic elution method had been appropriately used to symmetrically and well isolate colchicine, khellin, and their breakdown products. 60:40 mL of ethanol:filtered water make up the eluting system, which flows at 0.5 mL/min. The testing was detected at 245 nm, considering the concentration values of 3.5-24.5 mu g/mL for khellin and 1.0-7.0 mu g/mL for colchicine. With a total analysis time of <11 min., the chromatographic technique in dispute proved to be appropriate for regular colchicine and khellin quality control assessments. It made sense to add ethanol for the sake of getting a satisfactory resolution and being ecologically conscious. The impact on the environment was examined using three different methods: the eco-scale calculator, the AGREE, and the MoGAPI. The final AES result of 93, AGREE result of 0.68, MoGAPI result of 74, BAGI result of 80, and whiteness result of 97.5 all reveal how extremely ecologically conscious the chromatographic process is. Clearly, the concurrent tactic is accurate, precise, persistent, and ecologically sound. The perks and purity of the suggested approach were contrasted with those of the published method. Ultimately, both pure and sachet dosage forms of colchicine and khellin can be effectively employed for routine quality control analysis.
