Isolation, characterization of Enterococcus phages and their application in control of E. faecalis in milk

Abstract:
This study isolated and characterized twelve Enterococcus bacteriophages from raw milk, cow feces, compost, and poultry samples, and evaluated their use as a phage cocktail to control Enterococcus faecalis in milk. Among the isolated phages, PEF1, PEF7b, and PEF9 exhibited the broadest host ranges and strongest lytic activities and were combined into a three-phage cocktail. The cocktail showed superior antibacterial and biofilm removal activity compared with individual phages or two-phage combinations, reducing biofilm-associated cells by 6 log within 6 hours and remaining effective at refrigeration temperature (4°C). When applied to contaminated milk, the cocktail gradually reduced viable E. faecalis counts to below the detection limit after 48 hours at 4°C. These findings demonstrate that the developed phage cocktail is a promising natural biocontrol strategy for improving the microbiological safety and shelf life of dairy products.
