Characterization of selected phages for biocontrol of food‑spoilagepseudomonads

Abstract:
This study isolated and characterized bacteriophages targeting food-spoilage Pseudomonas species for application as biological control agents during refrigerated food storage. Twelve lytic phages were isolated from chicken and soil samples, and two phages, vB_PflP-PCS4 and vB_PflP-PCW2, were selected to formulate an optimized phage cocktail based on host range and lytic activity. The cocktail effectively controlled Pseudomonas fluorescens and Pseudomonas putida at 30°C, 8°C, and 4°C, significantly suppressing bacterial regrowth compared with individual phage treatments. Genome analysis showed that both phages belong to the Podoviridae family and lack tRNA, virulence, toxin, and antibiotic resistance genes. In addition, vB_PflP-PCS4 harbored a putative novel endolysin gene, highlighting the potential of this phage cocktail as a safe and effective biocontrol strategy for reducing psychrotrophic food-spoilage bacteria during cold storage and food distribution.
