Abstract:
Background and study aim: Currentschistosomiasis control strategies are mainlybased on chemotherapy, but manyresearchers believed that the best long-term strategy to control schistosomiasis isthrough immunization with antischistosomiasisvaccines. This study aimsat assessment of the efficacy of differentpotential anti-schistosomal vaccines (ascrude soluble egg antigens (SEA), solubleworm antigen preparation (SWAP) andcombined SEA & SWAP) byparasitological and molecular studies inexperimental murine models. Materials and Methods: Sixty malelaboratory bred Swiss Albino mice wereused and divided into six groups; controlnormal (G1), control infected by ± 80cercariae by S.C. route (G2), Freund’sadjuvant (adj.) received then infected(G3), SEA+adj. received then infected(G4), SWAP+ adj. received then infected(G5) and combined (SEA+SWAP) + adj.received then infected (G6). A scheduleof sensitization, immunization andschistosomiasis challenge were followedand performed on different mice groups.Mice were euthanized 10 weeks post-infection.Potential vaccine efficacy wasinvestigated by parasitological andmolecular studies including eggcount/gram stool using modified Kato thick smear, liver egg load, oogram pattern in the liver and stool PCR to detect S. mansoni egg DNA in stools ofstudied mice. Results: The combined (SEA+SWAP)vaccine caused the highest significantreduction in the fecal egg count followedby SWAP then SEA antigens. On the otherhand, the highest percentage reduction ineggs/gram liver tissue was attributed tothe combined (SEA+SWAP) followed bySEA then SWAP antigens. Regardingoogram results, the combined (SEA+SWAP) antigens were more efficient inincreasing the number of dead ova withhighly significant reduction in the numberof mature & immature ova, followed bySEA then SWAP antigens. The lowestpercentage of S. mansoni egg DNAdetected by PCR in stool samples wasencountered with the combined (SEA+SWAP), followed by SEA then SWAPantigens. Conclusion:The parasitological and PCR-basedassessment studies denoted that thecombined (SEA+SWAP) vaccine candidatewas the most effective in protectionagainst schistosomiasis challenge. Theresults of parasitological and molecularstudies were nearly similar but themolecular study was more sensitive, definite and accurate.
