Toxicity of Subacute Oral Zinc Oxide Nanoparticles on Testes and Prostate of Adult Albino Rats and Role of Recovery

Abstract:
Background: Zinc oxide nanoparticles (ZnO NPs) have immense potential and application in various types of cosmetic products and health care sectors.They have attractive properties, and attracted great interest worldwide.Yet despite its potentially interesting beneficial effects, it has been proven that they can be also cytotoxic and genotoxic for multiple types of human cells, especially the reproductive system. Aim of the work: Our study aimed to detect the effect of oral subacute ZnO NPs on the testes and prostate of adult albino rats and the role of recovery. Materials and Methods: Forty adult male albino rats were divided into three groups; control (positive and negative), ZnONP-treated group for four weeks and a similarly treated group left for supposed recovery improvement testing two weeks after cessation of ZnONP administration. All rats were examined for serum oxidative stress markers, testicular and prostatic tissue inflammatory cytokines, DNA fragmentation, and extended histopthological evaluation.Histological study was performed using hematoxylin and eosin and Masson trichrome stains. Immunohistochemical expression for iNOs was performed. Morphometric and statistical analysis were also performed. Results: Statistically significant difference in the oxidative stress and inflammatory markers and DNA shearing and aggressive histopathological testicular and prostatic changes were also detected in ZnONPtreated group as compared to the control and the follow up group that showed significant improvement.. Immunohistochemical examination of testicular and prostatic sections of ZnONP- treated group revealed highly statistically significant increase in the mean area percent of iNOs immunoreactions which significantly decreased in follow up groups. Conclusion: ZnONPs have detrimental male reproductive effects that can be controlled by limiting the exposure to such products.
