The evolving role of protein disulfide isomerase A3 in Egyptian bladder cancer patients

Abstract:
Background:ProteindisulfideisomeraseA3(PDIA3),anendoplasmicreticulumprotein,isexpressedinbladderofBCpatients.However,itsroleinBChasbeenelusivetillnow.Objectives:ThisstudywasconductedtoassesswhetherPDIA3geneexpressionwasassociatedwithincreasedoddsofBC,inparticularmuscle-invasiveBC(MIBC).Methods:Ninetythreepatientsunderwentcystoscopyandbladdertumorswerebiopsiedandhistologicallyas-sessed.Datacollectedincluding:patientdemographicsandclinicalcharacteristics.Biochemicalmarkers:hy-poxiainduciblefactor1-alpha(HIF-1α),interleukin6(IL-6),advancedoxidationproteinproducts(AOPP),Malondialdehyde(MDA),8-hydroxy2-deoxyguanosine(8-OHdG),andreducedglutathione(GSH)andmole-cularmarkerPDIA3geneexpressionweremeasured.Results:Accordingtothetumorgradeandstage,36patientswerefoundtohaveMIBC,27patientshavenonMIBC(NMIBC)and30patientshavenobladderlesions(controlgroup).PDIA3geneexpressionlevelhadthelargestcontributiontoamultivariablemodelforpredictingBC,whichachieved89.0%predictiveaccuracy.TheAUCfordiscriminatingMIBCsignificantlyincreasedfrom0.644to0.938whenbiochemicalmarkerswerere-placedbymolecularPDIA3markerinthefinalmodel.Conclusions:UsingPDIA3expressionalongwithpriorinformationofpatient'sage,bilharzialhistorywithgrosshematuria,canhelpclinicianspredictBC,discriminateMIBCanddecideconsequentlythemostpromisingtherapeuticmanagementinEgyptianpopulation.
