Some Histological and Ultrastructural studies on the testis of adult buffalo bulls (Bos Bubalis L.)

Abstract:
The present study was performed on 25 testes of adultbuffalo bull. For light microscopy the samples were fixed in 10%buffered neutral formalin and processed until sections of 5-7!lm inthickness were obtained and stained with different stains. Forelectron microscopy, the specimens were fixed in 3%glutaraldehyde solution in O.l M sodium phosphate buffer pH 7.4 at4°c for 3 hours and post fixed in 1% osmic acid. Sections werestained with uranyl acetate and lead citrate, examined andphotographed with with JEOL JEM-IOlO TEM operating at 80 kv.In Faculty of Medicine, Zagazig and Tanta Universities.The testicular weight of adult buffalo bull ranged from 225-250 gm, its width was of about 4-5 cm and its length of about 7-8.5cm. The testis of the buffalo bull was lain in scrotum with a verticallongitudinal axis, the head of epididymis was situated dorsally,while the tail was ventrally, The epididymal border of the testis waslocated caudally and the free border was craniallyThe histological finrlings:-Each testis was enclosed by white fibrous capsule; tunicaalbuginea, composed mainly of collagen fibers, the connectivetissues extend into the organ, forming a mass, called themediastinum testis. The seminiferous tubules were divided intotubuli contorti (convoluted seminiferous tubules) and tubuli recti(straight seminiferous tubules) which connect the tubuli contorti tothe rete testis. The lamina propria of seminiferous tubules waslimited by PAS positive basal lamina. Under EM the basal lamina
