Abstract:
Among most important diseases affecting cattle, and other animals and causing severe economic losses and is the Tuberculosis.This study was carried-out for :1. Isolation and Elimination of infected cattles through tuberclin test where the positive reactors go to abattoir for taking samples before and after slaughtering .2. Diagnosis of T.B in laboratory by different methods of bacteriological examination as culture on different suitable media, staining by Ziel-Nelson from tissues and positive culture .3. Evaluation by different advanced methods of identification as ELIZA, P.C.R. and flaw cytometer .4. Comparing of different tests aiming to use the more sensitive, accurate and Rapid method for isolation and identification of mycobacterium bovis Spp.5. This study was carried-out on one hundred and thousand (1100) cattle that, were undergo tuberculin test (Single intradermal) in cervical region.The test had done in two different places, the first is Motobos, Kafr El-Sheikh and the second is Kafr El-ziat city, along the period of August 2010 until March 2011. The positive reactors go to abattoir and slaughtered for more investigation by post-mortThis study concluded that :1. The most important factors affecting tuberculosis incidence were sex of the animal.2. The female cows of higher incidence than male. Also, the lesions were differ according to age , breed and system of management. The prevalence lesions were higher in old and young animals. Than middle ages and were prevalence in respiratory system, L. 3. The results of single intradermal tuberculin test in different cattle. showed that, the incidence of tuberculosis differ from herd to another herd according to the locality under study.4. Kafr El-sheikh governorate (Motobes region) of higher number of positive reactors observed at the age over 5 year. , followed by the age at 3- 5 years and the lower incidence observed at the age of 1- 3 years and still the kafr El-sheikh (Motobos regio5. The female dairy cows were highly affected than males in Kafr El-Sheikh .6. The post-mortem finding showed that the total positive reactors show visible lesions in 35 reactors by percentage 97.22 % from total positive reactors , while non visible lesions were 1 case by percentage 2.77 % from total positive reactors.7. The visible lesions were distributed mainly in head L.NS , bronchial L.NS, prescapular L. NS, mesenteric L. NS, heart, lung , liver, spleen and also the generalized T.B were observed.8. The lesions in all positive reactors were casiated nodules and the lower level of it calcified lesions.9. The results of cultivation on L.J with Sod. Pyurivate indicated that, 17 samples from visible lesion from different sites give positive growth for acid fast bacilli.10. The sample of non-visible lesion give negative results on L. J media with glycerol.11. The L. J media is the best media for isolation of tuberculosis.12. The results of direct smear stained by Z.N prepared from different sites of +ve lesions and from culture.The results of slides from +ve lesions were a total of 5 slides show a cid fast bacilli under microscope from total 35 +ve lesions samples by percentage of 17.28 %.13. ELISA test is one of the most important tests that gave accurate results for detection of tuberculosis.14. The results of L.C.D array indicated that spleen, L.N of mediastinal, spleen, heart gave a +ve results.15. We used a rapid immunochromatography assay for rapid detection of tuberculosis in infected animals.The results of immunochrmatographic assay cleared that :1. This test is rapid , accurate and rapid for detection of tuberculosis in cattle.2. The results also showed that, the reactor of cattle that gave a positive results gave a visible and non-visible lesions of TB in cattle .
