Clinicopathological studies on the effect of some hypoglycemic substances in rabbits

Abstract:
The present work was performed to study the effect of amaryl andpropolis in the treatment of diabetes mellitus and hyperglycemiainduced by using alloxan. Moreover the side effects of these drugswere studied.This study was carried out on one hundred New Zealandrabbits weighing 500 gm were divided to main 6 groups to study theantidiabetic activity and side effect of amaryl and propolis.Ten rabbitskept as - ve control (gp.l) .Sixty rabbits were divided into 3 groups(gps.2,3 and 4) to study the antidiabetic activity of drugs ,each groupcontaning 20 rabbits and all sixty rabbits in the three groups followinga 48h fast, were injected liP with alloxan at a dose of (50 mg / kgb.wt) freshly dissolved in sterile saline (50 mg/rnl),then injected INwith 10 ml of glucose 5% and kept on drinking water supplementedwith 10% glucose for the first 24 h after the alloxan injection .Within7 days after alloxan injection, diabetes mellitus was confmned by thedemonstration of hyperglycernia (blood glucose concentration wasmeasured using the glucose oxidase method). Animals with bloodglucose concentrations less than 250 mg/dl and died animals wereexcluded from the study.Diabetic rabbits in gp.(2) kept withouttreatment all over the experimental periods as +ve control, whilediabetic rabbits in gp.(3) were treated orally with amaryl by thetherapeutic dose for 2 months and diabetic rabbits in gp.( 4) weretreated orally with propolis by the dose 53.3 mg/kg b.wt for 2months.The remaining thirty rabbits were divided into equal 2 groups(gps.5 and 6) to study the side effect of drugs. Gp. (5) orally givenamaryl as gp. (3) and gp. (6) orally given propolis as gp.(4) .Drugswere given in gps.(3-6) using an esophageal tube.In all groups, blood
