Effect of indomethacin pregnancy and foetal development

Abstract:
SUMMARY:- Indomethacin has been proved to be the most valuable drug for analgesic, anti-inflammatory and antipyretic action .The present study was designed to evaluate the effect of indomethacin on pregnancy and foetal development. teratogenic effects :60 mature female rats were mated with 30 mate male rats . On the first day of gestation , detected by vaginal smears, the females were divided into 2 classes, each class contains 6 groups, each group contains 5 female albine rats . First class take therapGroups were as follow :-1st group : received no treatment and was considered as control group .2nd group : was administered indomethacin I.M ( 1.5 or 40 mg / kg b. wt / day ) form the 1st through 5th day of gestation3rd group : was administered indomethacin I.M ( 1.5 or 40 mg / kg b. wt / day ) form the 6th through 10th day of gestation .4th group : was administered indomethacin I.M ( 1.5 or 40 mg / kg b. wt / day ) from 11th through 15th day of gestation5th group : was administered indomethecin I.M (1.5 or 40 mg / kg b.wt / day ) from 16th through 20th day of gestation6th groups : was administered indomethacin I.M (1.5 or 40 mg / kg b.wt / day ) from the 1st through 20th day of gestation .On 20th day of gestation period, each dam was anaesthetized by using alcohol, chloralform, ether mixture (1:2:1). Then a caesarian section was performed to obtain the foeti and the uterine horns were examined for resorption sites . The foeti were examinedSkeletal malformations were absence of last coccygeal vertebrae, rudimentary sternbrae, fused ribs, absence of phalonges of fore and hind limbs .Liver section of dams administered indomethacin revealed single or multiple vacules inside cytoplasm of hepatic cells Kuffer cells, odema and leucocytic infilterations.Kidney section of dams given indomethacin displayed vacular and hyDROPic degeneration, congestion of renal blood vessels coagulative necrosis with pyknotic or absent nuclei .Heart sections of dams disclosed congested coronary blood vessels, Zenker’s necrosis was focally seen in myocardium . The wall of coronary arteries was thickened, vacuolated and hyalinized .Stomach sections of indormathacin treated dams revealed focal necrosis, erosions and ulceration in gastric mucosa.Placenta of indomethacin treated dams revealed focal area of coagulative necrosis in the placental villi, sever desquamation.ConclusionIn the light of the present study, it could be conclude that indomethacin is a teratogenic drug adversely affects maternal dams and foetal development producing morphological, visceral and skeletal malformations as well as histopathological changes .
