Abstract:
There are some fatal diseases facing newly born calves speciallythose of viral nature. Two major causative viral agents of these diseases are bovine coronavirus (BCV) and bovine rotavirus (BRV) which aremost familiar causes of neonatal calf diarrhea. Detection of these virusesin affected animals in addition to monitoring of the immune status ofcalves against such viruses requires the presence of available, effectiveKits of good quality and low cost. So the present work was planned toprepare diagnostic Kits for BCV and BRV in trials to provide localproducts usually in essential need .These locally prepared diagnostic Kits are saving time and high cost which could be needed to obtain similarimported products.The obtained experimental results revealed that: 1- The most suitable susceptible cell cultures for BCV and BRV were MA104 and MDBK yielding virus titers of7.5 and 8 10glO TC1Dso/ml forBCV and BRV, respectively. VERO cells were found to be lesssusceptible for both BCV and BRV showing virus titers of 6 and 6.5 10glOTCIDso/ml, respectively. While, BHK cells were found unsusceptible forpropagation of both viruses.2-It was successful to prepare anti-BCV and anti-BRV hyperimmune serain rabbits of good quality as shown by SNT, AGPT and estimation ofserum protein which revealed that the immunoglobulin contents were 6.4 . gldl and 10.5 gldl for BCV and BRV-antisera, respectively.3-Chekerboard ELISA indicated that the locally prepared whole cell andcell lysate antigens (CLA) of bovine BCV and BRV had optimal dilutionsof (1/30 &1150) and (1/40 &1150), respectively.
