Abstract:
irology. This work was carried out for molecular comparison between FMDV field strains causing recent outbreaks in Egyptand the used vaccine strains. 252 epithelial samples and 13 8oesophageal pharyngeal fluids were collected from 9 differentEgyptian governorates during the period of2009 to 2012.The result obtained from this study can be summarized as followings: .1. Isolation and identification of the FMDV current field strainsby tissue culture inoculation, baby mice infection and by R T-PCR using universal primer. The collected samples were isolated in BHK cell line forthree successive passages and also by baby mice inoculationobserved for seven days, all the suspected samples arepositive. The positive samples then retested by RT-PCR forgeneral detection of FMDV using universal primer, allsamples are positive.Typing of the isolated FMDV strains by CFT using specificguinea pig antisera to the 146S antigen of each of the sevenserotypes of FMDV, ELISA using specific rabbit antiserum asa trapping antibody, and specific guinea pig antisera as thedetecting antibody, and RT-PCR using RNA extraction byGeneJET™ RNA Purification Kit (Fermentas, EU), RT-PCRby Maxime” RT-PCR PreMix Kit (iNtRON, Koria) and five
