Studies on Different Histopathological Methods and Stains Used In The Demonstration of Fungal Elements In Tissue Sections

Abstract:
This work was carried out to study histopathology of most important fungi in man and animals namely Candida albicans,Cryptococcus neoformans, Aspergillus fumigatus, Aspergillus flavus,Aspergillus niger and Trichophyton mentagrophytes varmentagrophytes.Experimental infection were done intraperitoneally in rats withCandida albicans, Cryptococcus neoformans, Aspergillus niger andAspergillus flavus. On other hand Candida albicans infections wasalso done in rats and mice by oropharyngeal inoculation. Aspergillus.fumigatus was experimentally infected by intratracheally in rats whileTrichophyton mentagrophytes var mentagrophytes was experimentallyinoculated by scarification in the skin of rabbits. All animals subjectedto experimental infection were treated by cortisone before and after inoculation except in case of these infected by Trichophytonmentagrophytes var mentagrophytes. The animals were put underobservation of three weeks. Two animals were sacrified each weekand postmortem examination done then organs were taken for histopathological studies. In case of infection by Trichophyton mentagrophytes var mentagrophytes skin scrapings were collected tendays after inoculation. Specimen from organs of sacrified animalswith xylol then embedded in paraffin, paraffin sections of 51l thickness were obtained and stained with H&E and PAS.
