Abstract:
The present work was carried out on buffalo - semen to asses the post- thawing spermatozoal survivability following caffeine and heparin addition. Semen after being collected by artificial vagina was microscopically evaluated. Samples showed 600/0 motilitcitrate diluent containing 20% egg yolk, 2.9% sodium citrate , 500I.U. penicillin with 500mg streptomycin and glycerol which addedin 2 levels (Sand 7%), caffeine (2,6 and 10mM) was added to thediluent contain 5% and 70/0 glycerol level. Heparin (10 JL g) wasadded with the same concentrations, of caffeine at Sand 70/0 glycerollevels . Semen packed in 0.5 ml French straw and deep frozen inliquid nitrogen at -196 Oc . The post thawing spermatozoalsurvivability (motility, dead % and abnormalities ) was determinedThe best post- thawing spermatozoal survivability was used inA.I. of buffalo cows to detennin the pregnancy rate. This studyrevealed the following results,:l-Caffeine addition highly significantly stimulates the post-thawingmotility. The motility percentage was 44.7 ± 1.6 , 45.3 ± 2.44 ,38.8 ± 1.14 and 11.5 ± 1.43% for 0 ,2, 6 and 10 mM caffeine at7% glycerol and 40.5 ± 2.12,49.4 ± 2.50 , 40.3 ± 1.42 and 14.9± 1.29 % for 0 , 2 , 6 and 10 mM caffeine at 50/0 glycerol. Thehighest value was detected for 2mM compared with 6 and 10mMcaffeine in both glycerol levels.
