Abstract:
The present study was carried out on 1382 animals (533 Fresian cattle, 302 Native cattle and 547 buff-aloes) of a different sex, ages and at a various sources at Menofia governorate to clearify incidence of brucellosis and to find the most reliable or senIn this investigation using different serolo-gical tests reavealed that, Rose Bengal plate test showed higher positive reactors 89 (6.43% ) than standard tube agglutination test 62 (4.48%) or 2-mercepto-ethanol test 44 (2.89%).The results positive reactors in Rose Bengalplate test and 2-mercepto-ethanol test in this study agreed in 100% with those of complement fixation test whereas poSitive serum tube agglutination test agreed in 98.7%.Out of 105 cattle and buffaloes (reacted to tube agglutination test or Rose Bengal plate test or both) Application of Rose Bengal plate test on investi-gated animals at Monofia Governorate revealed, 72 (13.8%) reactors among fresian cattle, 3 (0.99%) for Concerning the effect of age and sex on the result of diagnositic tests of examined animals, the results of Rose Bengal plate test showed, 86 (7.74%) reactors among adult animals [77 (9.62%) in cows and 9 (2.90%) in bulls ] and 3 (1_10%) in young one 11 (The results of serum agglutination test when compared with those of semen agglutination test of 220 bulls, revealed that 20 (9.09%) bulls were reacted to serum agglutination test, usually at a lower titre (1/10 and 1/20), while the semen plasma agglutinatBacterological examination of semen from reacting bulls failed to recover brucella organisms. CONCLUSIONfrom the results of the present study, it is concluded that:Rose Bengal plate test is an effective diagnostic test for bovine brucellosis as it dectects more positive reactors than standard tube agglutination test or 2-mercepto-etharol test. Beside this it is easy and economic test for screening of herds.No single serological test was absolutaly accurate for diagnosis of bovine brucellosis, and all of them detect reactor animals but with a relative sensitivity Sensitivity of both Rose Bengal plate and complement fixation test are still superior than otherPrevalence of brucellosis in Monofia Governorate ishigher in fresian cattle than in buffaloes or native cattle, higher also in adult cattle than young ones, higher among cows than in bulls, and in farms than in individually bred cattle in villages, and among cattle with reproductive troubles than in apparBlood serum tube agglutination test detects more positive reactors than Milk ring test, but Milk ring test is much usefel than other tests for early detection of the reactors in herds and much applicable from the practical point of view.Bulls positively in serum tube agglutination test do not usually reacting to semen agglutination test.
