Abstract:
Toxicological and immunosuppressive effects of ”5-aza-cytidine” were studiedin order to investigate some aspectsof one of this newly antitumoroginic agents which is used asa cancerostatic drug in human leukemia. During our studiesthe following experiments were carried out:1- The TD50 and 1D50 of ”5-azacytidinetl on monkey stablecell line were determined and found to be 200 ”g/ml and230.67 ! g/ml of growth mediu..rn respectively.12. 1t5-Azacytidinel1 in different concentrations produceshistopat~ological effects on M.S. ~ll line manifestedby cellular death and lysis of t:i1e cytoplasm manifestedby deeply stained nuclei or presence of cellular vaculesin the cytoplasm.3- Alkaline phosphatase enzyme activity VffiS jncreased spec-ially at 50 ’!g/ml dose while acid phosphatase was at itshighest activity at 5 ,;g/ml dose.4- Fat and cellular glycogen content were not investigatedby our technique used.5- RNA and DNA cellular contents were increased at lowerdose level ( 0.5 • 0.67 ~nd 19/ml of the medium). HigherconcentratioE { 50 and 200 !;g/ml of medium) producedlower HHA and D~TA macromolecules in M. S. cell line.6- Mitotic index of IvI. S. cell line was lowered of both higherand lower dose levels and this lowering was dose related.
