Studies On The Tissue Culture Rabies Vaccine

In this study, trials were made for the adaptation of two rabies virus stains: The Flury LEP and the CVS, in three Cell lines (BHK, VERa and MS cells) and in the primary chicken embryo fibroblasts. Inthe propagation of virus in cell lines, three differentTwo methods were used for propagation of virus inchicken embryo cells: adsorption and virus cell mixingtechniques. The only method that proved to be successfull in propagation of rabies virus in BHK cells was thealternative method. The virus titre after gradual increase, reached up to 106 in the lOth passage. By using the direct immunofluorecent antibodytechnique the virus antigen content in the infected cells was detected. It gradually increased in the cells during the serial passages and reached its maximum from the 8th to the lOth passage when nearly 80-90% of cells showed flourescence.
