Enhancement of Release for certain anti-Inflammatory drugs and evaluation of their permeability through the skin

ABSTRACTNon-steroidal anti-inflammatory drugs (NSAIDs) are among the most frequently prescribed drug groups. These drugs are used dermally or systemically in treatment of different rheumatic diseases, including rheumatoid arthritis, osteoarthritis, low back pain and some joint diseases.At present, oral administration is the most common form of drug delivery for such drugs. Oral route has the advantage of easy administration, however it has significant drawbacks namely poor bioavailability due to hepatic metabolism, therefore, the need for high or frequent dosing are required, which leads to stomach pain and vomiting.Topical drug delivery system is an alternative way to overcome the disadvantages of oral route. Topical drug delivery systems allow localized administration of the drug anywhere in the body through ophthalmic, vaginal, skin and rectal routes. Topical formulations encompass a wide variety of formulations intended for cosmetic or dermatological application, to healthy as well as diseased skin. Topical preparations can be formulated with varying physicochemical properties, as solid, semisolid or liquid.Recently, transdermal drug delivery systems have been extensively studied, since it offers several important advantages over other traditional dosage forms. They offer steady permeation of drug across the skin allows for more consistent serum drug levels which is often a goal of therapy with more patient compliance.
