Synthesis of certain Thienopyrimidine of Pharmaceutical interest

Abstract:
Research Objective Thieno [2, 3-d] pyrimidines with a wide spectrum of biological activities are known. Several of these derivatives possess potent analgesic- anti inflammatory(2,4), cardiac (10-14) , anticancer (6-9) antibacterial(15) antiviral (17), antiallergic(21 ,22 ) , uricosuric (22, 23) anticonvulsant (1) and antibelharzial (26)activities .Since certain derivatives of pyrazolines were reprted to possess potent anti inflammatory activity (71, 72), it deemed of interest to prepare compounds having the general formula VIa-d, VIIa-d and VIIIa-b with the aim of having anti-inflammatory activity.On the other hand, many of pyridone derivatives have useful applications as anti-inflammatory (73, 74, 75), hypnotic (76) and some are used in skin diseases (77). Also, aminopyridines are reported to have excellent anti inflammatory activity (78, 79, 80, 81). So, it was of interest to synthesize novel thieno pyrimidines XIIIa-c and XIVa-c in conjunction with such derivatives with the objective that the new compounds might show promising anti-inflammatory as well as cardiovascular activity. Certain aminothiazoles were reported to possess anti inflammatory (82), anti tubercular (83) activities, and others are used to promote the wound healing (84) compound XV was constructed with the hope possessing anti-inflammatory or antituberculotic activities.It was reported that substituted semicarbazones and thiosemicarbazones have been evaluated as anticonvulsants, local anaethtices, antiarrhythmics, and antimicrobial agents (85). Moreover, varioues pharmacological properties have been shown to be associated with thiazolidinones, thiazole derivatives, including anti-inflammatory (86, 87) anticonvulsant (88, 89), antimicrobial (90, 91, 92) and antitubercular activity (93). On the light of the above findings , the incorporation of the semicarbazone , thiosemicarbozone , thiazolidinone , thiazoline and imidazolidinone moieties to thienopyrimidine substrate was suggested for creating new compounds XVII, XVIII, XIX , XX and XXI of possible anti-inflammatory , anticonvulsant , antimicrobial or antitubercular properties.
