The effect of some Anti-infammatory drugs on the Mediators of Inflammation

Abstract:
The present study was designed to investigate the influence of two non-steroirlal anti-inflammatory drugs namely, indomethacin and piroxicam on certain pharma¬cological aspects of some mt”diatorS of inflammation. Indomethacin was used as a stand~rd drug while piroxicam was used as a structurally rlovel NSAID. Both drugs are potent inhibitors of endogenous prostaglandin biosyn¬thesis.The inflammatory mediators chosen included histamine, serotonin and prostaglandins. The release of both histamine and prostaglandins from the isolated perfused guinea-pig lungs was investigated in sensitized animals under the influence of the two NSAIDs. Both indomethacin and piroxicam inhibited the endogenous release of prostaglandins from the sensitized perfused lungs to approximately the same extent during antigen challenge, while histamine release was not altered by either drug.
