Abstract:
Analysis of pharmaceutical amines is required for purposes of quality control of drugs and their dosage forms, for drug stability studies, as well as for study of drug absorption and pharmacokinetics. There is a growing need for simple and specific methods for the analysis of such drugs.from the medicinal point of view tertiary amines including nitrogen heterocyclic constitute an important group of pharmacologically active agents which in fact is the majority group when compared with secondary or primary amine drugs.Photometric methods are considered simple, sensitive and time saving in comparison with other techniques, particularly the classical stoichiometric methods which usually lack specificities, may be time-consuming, sometimes exhibit difficulties in detecting the end point, in addition they often require large quantity from the sample.For pharmaceutical amines in general very few direct ultraviolet methods are available owing to the inherently low molecular absorptivities of the majority of these compounds. Furthermore compared to secondary and primary amines there are few really good colorimetric methods for tertiary amines. This is because of the lack of hydrogen atoms of the nitrogen of the tertiary compounds and hence a narrower range of possible chemical modification leading to formation of chromogens i.e. tertiary amines have fewer analytical ”handles”. Thus whereas a variety of derivatives is possible for secondary and primary amines (carboxyamides, sulfonamides, nucleophilic substitution on chromophore rich arene…etc.) tertiary amines are not amenable to such derivative formation. Consequently additional sensitive photometric methods for pharmaceutical tertiary amines are still wanting.
