The objective of this work is to formulate Buflomedil hydrochloride fast soluble tablets. The work in this thesis was divided into three main chapters:Chapter I: Formulation and evaluation of Buflomedil hydrochloride fast soluble tabletsChapter II: Stability study for selected Buflomedil hydrochloride fast soluble tabletsChapter III: Bioavailability and pharmacokinetic studies of selected Buflomedil hydrochloride fast soluble Formulation and evaluation of Buflomedil hydrochloride fast soluble tablets .Three different methods were selected for preparing Buflomedil hydrochloride fast soluble tablets; these methods are direct compression method (DCM), blister tablet method (BTM), and freeze drying method (FDM).Different fillers (Lactose fast flow, lactose monohydrate, and mannitol), four types of disintegrants used for fast disintegration at two different concentrations (croscarmellose, sodium starch glycolate, pregelatinized starch, and Avicel PH101), different binders at different two concentrations (PVP K25, PVP K 30, PVP K90), talc powder as lubricant, and Buflomedil hydrochloride (150 mg / tablet) were used in this study.Thirty two different Buflomedil hydrochloride fast soluble formulae were prepared by direct compression method (DCM).The composition of these formulae as follows: F# 1 to F# 8 are composed of lactose fast flow as filler, croscarmellose [2, 5 %], Avicel PH101 [10, 20 %], and polyvinyl pyrrolidones (PVP) K30 [3, 5 %], F# 9 – F# 16 are composed of lactose fast flow as filler, sodium starch glycolate [1, 3 %], Avicel PH101 [10, 20 %], and polyvinyl pyrrolidones (PVP) K25 [3, 5 %], F# 17 – F# 24 are composed of mannitol as filler, croscarmellose [2, 5 %], pregelatinized starch [5, 15 %], and polyvinyl pyrrolidones (PVP) K30 [3, 5 %], and F# 25 – F# 32 are composed of lactose monohydrate as filler, pregelatinized starch [5, 15 %], Avicel PH101 [10, 20 %], and polyvinyl pyrrolidones (PVP) K90 [3, 5 %].The prepared medicated fast soluble tablets were evaluated for Buflomedil hydrochloride content, weight uniformity, friability, and hardness.
