Abstract:
AbstractThe intestinal absorption of oral antidiabetic drugs such as metformin which used in the treatment of type-II diabetes is altered when concomitantly administered with antacids, antinuclear agents, antibiotics and others. The absolute bioavailability of a metformin given under fasting conditions is approximately 29-60 %, pharmacokinetic interactions may also increase the adverse effects of metformin. Recently, the clinician and clinical pharmacologist are focusing their attention to report and manage these interactions for the better control of type II diabetes mellitus with metformin.So, we aimed to enhance metformin bioavailability by using different ratios of two polymers eudragit RS100 and ethyl cellulose in the form of nanoparticles.Also, a randomized parallel studies in one phase was carried out to evaluate the bioavailability as well as the pharmacokinetic profile of metformin hydrochloride administered with three antibiotics namely: amoxicillin, cefadroxil and ciprofloxacin.After formulation and characterization of metformin as a nanoparticles , the best formula which composed of metformin and eudragit RS100 (1:3) was chosen to compared with metformin brand drug (Glucophage).This best formula was feed with amoxicillin to evaluate the bioavailability and the interaction between amoxicillin and metformin.
