Interaction of Anti-Inflammatory Agents with Oral Antidiabetic on Certain Metabolic Aspects in Experimental Animals

Abstract:
The effect of adjuvant arthritis and its treatment with NSAI drugs (phenylbutazona and diclofenae sodium) on hepatic carbohydrate and lipid metabolism as well as plasma corticosteroids was studied. Furthermore, the study was extended to include the combined effect of the above drugs with the oral ant diabetic (tolbutamide) on the same metabolic aspects . Adjuvant arthritis was induced in rats by administration of complete Freund’s adjuvant as a single intradermal injection of 0.1 ml of the adjuvant in the palmar surface of the right hind foot pad . On the 15th day of adjucant inoculation m the arthritic rate were divided into several groups . These groups received oral doses of either phenylbutazone (50 mg/kgm) and combination of tolbutsmide with either phenylbutazone ( 200 mg + 50 mg / kgm) or diclofenac sodium ( 200 mg + 2 mg / kgm) . Analysis was done 2 hours after drug administration for studying the acute effect of the drugs . Other groups of arthritic rats received daily oral doses of the anti-inflammatory drugs either separately or in combination with tolbutemide as previously motioned but for a period of 7 , 14 and 21 days treatment . Chronic effect of these drugs was studied 24 hours after the last dose administered . Mean while control groups of normal amd arthiritic rats were sacrificed on the 15th , 22nd , 29th and 36th day of adjuvant inoculation . Liver glycogen , glucose – 6 – phosphates , phosphorylaes as well as by evaluating blood glucose level . The metabolic pattern of liver lipids was studied by evaluating liver cholesterol , cholesterol esters, triglycerides , non esterifies fatty acids , phospholipids and total lipids .
