Synthesis of Certain substituted Hydantoins & Thiohydentoins of Pharmaceutical Interest

Abstract:
A survey covering the synthesis as well as the chemical and biological properties of hydantion and thiohydantoin derives is included .Abrief account summarizing the importance of arylidene ,hydrazono and Mannich –type bases as anti- microbial agents is given .The different methods of preparing the inter –Mediates are thoroughly investigated . The experimental results attained are critically discussed .The present investigation comprises the prepara- tion of the following intermediates :3- Methy 1-2,4- imidazolidinedioneII. 1,3- Dimethy 1-2,4- imidazolidinedioneIII. 3- Methy1-2- thioxo -4 – imidazolidinoneiV. 1,3- Dimethy1-2- thioxo -4- imidazolidinoneThe work involves the condensation of these key compounds with different aromatic aldehydes to afford the corresponding 5- arylidene derivatives .Attempts to couple methl and dimethlhdantoins wish aryl diazoniumsalts were unsuccessful , whereas the sulphur analogues gave the expected 5- arylhydra zono derivatives . Trials of their desulphurization , in alkaline medium , resulted in rearrangement of the thione- carboxic acid.Meanwhile, the sulphurated intermedidates were successfully condensed with different secondary amines and formal dehyde to produce the corresponding Mannich- type bases at c5.
