Chemical and Biochemical Modifications of Paeoniflorin and Major Metabolite Paeonimetabolin-1,targeting Nuropharmacolgical Activities

Abstract:
The strummed dissertation is prefaced by a comprehensive literature survey on peony roots, their pharmacological activities and chemical constituents. The thesis also concerned with the pharmacological activities and the intestinal bacterial metabolism of paeoniflorin, the most important constituent of paeony roots. In addition to the anticonvulsant activity of its major metabolite paeonimetabolin-I. The introduction included also a study bout the pharmacokinetics oof paeoniflorin and paeonimetabolin-I. Moreover the thesis include a literature survey on the Alzheimer’s disease, epilepsy and the drugs used for treatment of these diseases.The current investigation involved the following steps :• Isolation of paeoniflorin from paeony roots.• Chemical and biochemical modifications of paeoniflorin and paeonimetabolin-I.• Evaluation of the antiamnesic and/or anticonvulsant activities of the designed compounds.• Resolution or R and S isomers of the most active thiopaeonimetabolin-I derivatives and evaluation of their anticonvulsant activity.• Isolation and characterization of novel paeonilactone-A derivatives.Assignment of the chemical structures of the newly prepared compounds was based upon elemental analyses, IR, MS as well as 1H, 13C-NMR spectral data and sometimes 2D-NMR techniques (NOESY and COSY spectra). On tee other hand, the reported compounds denoted by (*), were confirmed by matching their melting points with that reported as well as IR, MS and 1H, 13C-NMR spectral data.
