Background: The orexins (A and B), also termed hypocretins, are hypothalamic neuropeptides initially involved in the control of feeding behavior. Concerning the reproductive system, the functions of orexins seem to be multifaceted and likely to involve a complex array of central and peripheral actions.Objective: The present study was designed to investigate the effect of orexin- A on: 1.Circulating levels of gonadtotropins (FSH and LH) in intact and gonadal steroid unprimed and primed ovariectomizd (OVX) female adult rats. 2 .Gonadal functions. Moreover, the possible mechanisms of orexin- A action.Material & Methods: The present work had been carried out on 72 adult female albino rats , divided into I. OVX group : rats were castrated and divided into seven equal groups (n=8 rats in each one) : control OVX, orexin-A treated- OVX, steroid {estradiol (E) and progesterone (P)} treated- OVX, E+P with orexin-A treated OVX, CRFR1 blocker (Antalarmin) with orexin treated- OVX, CRFR2 blocker (Asstressin 2B) with orexin treated- OVX, and opioid receptor blocker (naloxone) with orexin treated- OVX groups, In all orexin treated groups, orexin A injected intravenously (i.v) in a single bolus dose of 50 g/Kg. BW. II. Intact group: subdivided into two equal subgroups (n=8) 1. control and 2. chronic orexin treated group: rats were injected daily with orexin-A i.v at a dose of 50 g/Kg. BW for 9 days. Blood samples were collected, serum was separated and LH, FSH, estradiol and progesterone levels were measured, also ovaries were isolated from control intact and chronic drug treated groups for histopathological studies.
