Abstract:
midazolam is a water-soluble benzodiazepine characteri-sed by two main advantages over other benzodiazepines: Exce-llent local tolerance and rapid onset and short duration ofaction. This thesis was planned to be a combined experimentaland clinical study of this new intravenous anaesthetic aqentin an attempt to elucidate its main properties and to evalu-ate its real place in anaesthetic Dractice.1- Experimental studies:A- In vivo experiments:a) Anaesthetic activity and acute toxicitystudiesfollowing its i.v injection in mice (determinationof ADSO and LDSO).b) Effect of different increasinq doses of the drug onECG, arterial blood pressure and respiration in dogsanaesthetised with chloralose.In vitro experiments:a)Effect of different increasing doses of the drug onthe contractions of isolated perfused toad’s heart.
